I was informed that my passport has been locked up in your office by Kumasi passport office directors (IMMIGRATION OFFICERS).

Your passport isn't locked up anywhere here, we don't have an office and are in no way connected to any immigration for any country

City officials do not do passports. They do not lock a person passport up, they will either issue a passport or reject a application. You need to go in and speak to immigration official directly.

Normally if there is no issue, a Ghana passport is issued within two weeks.
