Is the dishwasher overflow vent on the sink required?
I need to install a water purifier faucet and that location would be great.

Yes it's needed. It's not a vent but a Air -Gap or "Indirect Waste".
However, I can show you a way to use the counter top hole install your faucet and still provide a air gap for the DW.
A high loop, (see image) will take the place of a counter top air gap and , in my estimation, do a better job with no floods or maintenance.
Purchase a dish washer hose long enough to loop up all the way to the top of the cabinet and connect back to the disposal unit. Secure the loop with a pipe strap or simply wire it up. Although we have had many complaints with counter top air gaps we have yet to receive the first complaint on a high loop air gap. Good luck, Tom
