5 seconds is a very good time for hot water to arrive to upstairs bath. What exactly is the problem ?

Water system is a closed, pressurized, system, that is constantly under pressure. Air Lock will happen during repairs ( when heater is drained or section of pipe is replaced ) but should disappear first time you open faucet. Once you "bleed" the pipe of air, you should be getting water right away

If you think you have an air lock in hot water, go to the most distant faucet in your house, open it, and let it run for few minutes. If multi-level residence, you may have to do it on each floor.

Few questions: Is that a farm house ? Are you on well water ? Are you on booster pump ? Do you have hot water circulating pump ? Does the same problem occurs in other faucets in that bathroom and on the same level in other faucets ? Is that family house or apartment building ? What kind of water heater do you have ? What kind of faucet is it ?
