I'm remodeling a small, basement bathroom, and I want to remove sink and toilet first. However, only the cold-water line has it's own turn-off valve. To turn the hot-water line off, I have to turn off the hot water to our primary bathroom directly above it on the first floor. Can I temporarily cap the hot water line, then turn the hot water back on, while I'm remodeling the small basement bathroom? If so, what do I use to cap it?

You could add a shutoff valve.

You could use a Sharkbite Union with a small piece of pipe and a cap. The Sharkbite is removeable with a small tool.

You can also use a push on push off shut off valve.

Or you can sweat on a cap.

I would personally just add a shut off, will solve a lot of problems in the future

Fr Chuck raises a good point... ;)

However, I would actually cap both the hot and the cold water lines with sharkbite caps and then after the bathroom is remodeled and it is time to install the vanity and faucet Richard can install new chrome compression angle stops and new chrome cover plates for a nicer result... see images (other shutoff is probably old and needs replacing anyway).

These angle stops do not require soldering.

If the pipes are not copper pipes the same advice still holds but you will be using threaded caps and shutoffs instead.

If you decide to install shut off valves as Mark suggested in his post above, I will also say that you can install brass craft quick connect 1/4 turn angle valves they are easy to install and no tools needed, you can find them in home depot, them, see the images below.
