Can I take the wire 10/3 wire that comes from my well pump -- put a plug on it and run it off my portable generator

You will need a straight 240 plug, an outlet on the generator and the capacity in the generator to power the pump. You should install a grounding rod since the ground line on the pump motor is only bonded to the gen frame, not to ground.
