I want to disconnect the supply line near the toilet tank, and cannot unscrew it by hand, it does not seem to be in the shape (rounded nut) that a adjustable wrench could work , what should I do?

Attack it with a pair of pliers. Good luck, Tom
Thanks - seems like I need to get a pair of big pilers to fit the size, should I look for any specific pilers?

Even a pair of slip joint should work.
More commonly known as channel locks. I always call them pump pliers too. Channel locks is just a brand name.
