What's better to build a room in a basement with wood studs or metal studs
I have done both. Metal studding is faster and easier. Don't have to cut stud exact size. Do not have to secure studs in place and can adjust for s/r easier.
Would recommend frame door opening and window opening with wood so u can install trim with finish nails.

If the basement is dry both are fine. The floor plate should be made from pressure treated lumber. If the basement is damp you should consider way to keep it dry before building any room down there
Personally I would go with wood... hands down choice. I agree with ballenger with the pressure treated bottom plate but I'd take it a step further. I would also put down a thin foam membrane to separate the bottom plate and the concrete floor. Is it overkill? Yes. But then you should never have to worry about it. True, metal is faster but wood will also be better for hanging things on your wall.
On another note, I would leave a 1 inch gap where the sheetrock meets the floor just in case there are ever any water issues. The gap will be covered by the base molding. Even if you don't have any moisture problems, you never know if a pipe or water heater will break.
