There are scientific testing options other than "DNA testing", if that's what you are asking.
And of course if the man knows he didn't have sex with the mother within the pertinent time-frame, or if it can be proven he was sterile at the time, those would tend to prove it too.

One situation which has been known to happen is this: the child has a medical condition which requires his or her nearest relative to donate blood, bone marrow, etc. Tests are performed and they happen to indicate that the "father" is not the biological father after all.
You might look into what testing is available. It may be possible, for example, to have a test performed with DNA specimens gathered without your husband's knowledge. You do know, don't you, that these tests are usually done with cheek-swab samples?
Yes there could be many situations where your son's or your husband's medical condition might require testing that could determine that he's not the father.
Certain blood types are incompatible. So just blood typing could tell.
To add to what scottgem said, some eye colors are incompatible. Its not always easy to tell on a baby because eye color changes so much as they get older, but generally speaking, two people with recessive colored eyes (blue for example) don't have brown eyed babies.
