    HPLC is used to separate compounds based on their physical properties. For example, if you wanted to separate compounds based on their polarity, you would know (assuming a non-polar stationary phase and polar liquid phase) that the compounds that come off the column first would be the most polar and those coming off later would be less and less polar. LCMS works on a similar principle but the scale is much smaller and a mass spectrometer is used. With respect to your question, "fractionation" refers to the separation of compounds based on their physical properties as described above. Please keep in mind that this description is an extremely simplified explanation of how these analytical techniques work.
