I was wondering if anyone could help me?

I am American-Greek, I was born here is America. My Husband and I are thinking about moving to Greece. My grandfather is 100% greek;however, he was not born in Greece. His Mother, my great-grandmother was born there, his father was to. Can I become a citizen with proof that they were born there, and I also have other relatives still residing there. Can anyone help?

If your paternal grandfather's parents were born in Greece, and they are registered in the municipal records in Greece, then you should be fine. The process will take a few years, and is complicated [you need to find your parents' and grandparents' wedding certificates, and birth certificates] but you should be okay.

If it is your maternal grandfather's parents, then it is a bit more complicated.

The key thing is to find the municipal registration of the ancestor who was born in Greece. Without that, it doesn't matter that anyone is "100% Greek" - it will not matter to the authorities... So find the city/village where the ancestor was born, and see if they are registered in the "Dimotologio" or the "Mitroo Arrenon". Either of the two will get you what you need.
