I hold a G4 visa and will marry my girlfriend soon. She has been living illegally in the US for more than 5 years now.

If I marry her and apply for a G4 dependent visa for her, if the visa was granted and she goes home to our home country to get the visa, will she be able to enter the US or would she be denied at the port of entry?

Her being overstayed will trigger the 10 years bar and she won't be eligible for any visa. When you apply for G-4 dependent visa, you will need to file a waiver. It's recommended to consult immigration attorney.
