When wiring a 100 amp sub panel do you have to run a neutral and a ground wire?

Yes! You also need to know the distance between the main panel, whether you will be using above ground cables, below ground cables.

Is the sub panel going to be in the same building or a separate building? If a separate building, you will need a separate grounding rod and, Neutral must be isolated from Ground within a sub panel.
