Can work question employees regarding handicapped parking and placards?

If you have a handicapped placard and are parking in their handicapped spot then I think they have a right to ask about your disability. They are required by law to make reasonable accommodations to handicapped individuals to comply with ADA, and they are not out to get rid of you.
