Can I run main floor toilet drain into basement ejection pump pit.

Sure, but I'd like to know why and why only the toilet? Also, what fixtures are on the ejector pit now?

My first thoughts are that you will probably need to revent the toilet and connect it into the pit through the inlet on the side of the ejector pit. This means digging some of the floor surrounding the ejector pit up.

Otherwise, if you tried to dump in from the top of the ejector, through the lid, you could have trouble with stuff getting hung up on the ejector float and I also see issues getting a gas-tight seal.
